Kisspeptin is a neuropeptide encoded by the KISS1 gene and produced primarily in the hypothalamus. It is a master regulator of the reproductive hormone axis — acting as the primary upstream signal for gonadotrophin-releasing hormone (GnRH) release, which in turn governs LH, FSH, testosterone, oestrogen, and the full reproductive endocrine cascade.

Mechanism of Action

Kisspeptin binds the GPR54 receptor (also known as KISS1R) on GnRH neurons in the hypothalamus, triggering pulsatile GnRH release. This pulse-driven GnRH signal is the upstream driver of the entire HPG axis (Hypothalamic-Pituitary-Gonadal axis). Without sufficient kisspeptin signalling, the HPG axis cannot function optimally — regardless of how healthy the pituitary or gonads are. This makes kisspeptin directly relevant to any condition of HPG axis insufficiency, including hypogonadism with central aetiology.
