Sermorelin, GHRP-6 and GHRP-2: Dual-Axis GH Stimulation Explained
The combination of Sermorelin with GHRP-6 and GHRP-2 is one of the original multi-peptide GH-axis protocols — predating the Ipamorelin era and forming part of the foundational evidence base for GH secretagogue therapy.

The combination of Sermorelin with GHRP-6 and GHRP-2 is one of the original multi-peptide GH-axis protocols — predating the current Ipamorelin era and forming part of the foundational clinical evidence base for GH secretagogue therapy. It combines a GHRH analogue with two distinct ghrelin-receptor agonists, creating three simultaneous inputs into the GH secretion pathway.
What Each Compound Contributes
Sermorelin provides the GHRH signal — binding the pituitary GHRH receptor and triggering somatotroph transcription and GH release. GHRP-6 (Growth Hormone Releasing Peptide 6) adds a ghrelin-receptor stimulus with a secondary effect on appetite via central ghrelin pathways — useful for patients in caloric deficit or requiring anabolic support. GHRP-2 provides a cleaner ghrelin-receptor stimulus with slightly stronger GH pulse amplitude but more impact on cortisol and prolactin than GHRP-6.
Clinical Profile
The triple combination produces a larger and more sustained GH pulse than Sermorelin alone, with the GHRP components contributing both amplitude enhancement and receptor sensitisation. It is an older protocol, with many practitioners now preferring the Sermorelin and Ipamorelin or CJC-1295 and Ipamorelin combinations for their improved side-effect profiles. The Sermorelin/GHRP blend remains clinically relevant for patients where appetite stimulation or specific pulse kinetics are desired.
- Three simultaneous inputs into the GH secretion pathway
- GHRP-6 adds appetite stimulation — relevant in caloric restriction protocols
- GHRP-2 provides stronger GH pulse with some cortisol/prolactin elevation
- Ipamorelin-based combinations now generally preferred for side-effect profile
This combination is prescribed when the clinical profile of GHRP-6 or GHRP-2 is specifically indicated. Our physicians will assess whether this blend or a more modern combination better fits your protocol. Book a consultation at /book.
